چکیده

1 Knowledge about the functional status of the frontal cortex in infancy is limited. This study 2 investigated the effects of polymorphisms in four dopamine system genes on performance in 3 a task developed to assess such functioning, the Freeze-Frame task, at 9 months of age. 4 Polymorphisms in the catechol-O-methyltransferase (COMT) and the dopamine D4 receptor 5 (DRD4) genes are likely to impact directly on the functioning of the frontal cortex, while 6 polymorphisms in the dopamine D2 receptor (DRD2) and dopamine transporter (DAT1) 7 genes might influence frontal cortex functioning indirectly via strong fronto-striatal 8 connections. A significant effect of the COMT ValMet polymorphism was found. Infants 9 with the Met/Met genotype were significantly less distractible than infants with the Val/Val 10 genotype in Freeze-Frame trials presenting an engaging central stimulus. In addition, there 11 was an interaction with the DAT1 3’ VNTR polymorphism; the COMT effect was only 12 present in infants who did not have two copies of the DAT1 10-repeat allele. These findings 13 indicate that dopaminergic polymorphisms already affect selective aspects of attention in 14 infancy, and further validate the Freeze-Frame task as a frontal cortex task. 15 16

Genetic associations with reflexive visual attention in infancy and childhood.

This study elucidates genetic influences on reflexive (as opposed to sustained) attention in children (aged 9-16 years; N = 332) who previously participated as infants in visual attention studies using orienting to a moving bar (Dannemiller, 2004). We investigated genetic associations with reflexive attention measures in infancy and childhood in the same group of children. The genetic markers (...

The effects of child maltreatment and polymorphisms of the serotonin transporter and dopamine D4 receptor genes on infant attachment and intervention efficacy.

This investigation examined the extent to which polymorphisms of the serotonin transporter linked promoter region (5-HTTLPR) and the dopamine receptor D4 (DRD4) genes differentially influenced the development of attachment security and disorganization in maltreated and nonmaltreated infants at age 13 months, and the extent to which the efficacy of preventive interventions to promote attachment ...

Primary Dissociated Midbrain Dopamine Cell Cultures from Rodent Neonates

The ability to create primary cell cultures of dopamine neurons allows for the study of the presynaptic characteristics of dopamine neurons in isolation from systemic input from elsewhere in the brain. In our lab, we use these neurons to assess dopamine release kinetics using carbon fiber amperometry, as well as expression levels of dopamine related genes and proteins using quantitative PCR and...

Association studies of WD repeat domain 3 and chitobiosyldiphosphodolichol beta-mannosyltransferase genes with schizophrenia in a Japanese population

Schizophrenia and schizophrenia-like symptoms induced by the dopamine agonists and N-methyl-D aspartate type glutamate receptor antagonists occur only after the adolescent period. Similarly, animal models of schizophrenia by these drugs are also induced after the critical period around postnatal week three. Based upon the development-dependent onsets of these psychotomimetic effects, by using a...

Common variants in psychiatric risk genes predict brain structure at birth.

Studies in adolescents and adults have demonstrated that polymorphisms in putative psychiatric risk genes are associated with differences in brain structure, but cannot address when in development these relationships arise. To determine if common genetic variants in disrupted-in-schizophrenia-1 (DISC1; rs821616 and rs6675281), catechol-O-methyltransferase (COMT; rs4680), neuregulin 1 (NRG1; rs3...
